There are no nucleus in prokaryotes, therefore both transcription and translation occur in the cytoplasm. In eukaryotes, once transcription is over, mature mRNA leaves the nucleus through nuclear pores and goes to ribosomes in the cytoplasm for translation. Ribosomes then read the genetic code in the mRNA sequence to produce a polypeptide chain. Ribosome consists of large subunit and small subunit. The subunits are measured in Svedberg units (S). Prokaryotic ribosome has small (30S) subunit and large (50S) subunit. Eukaryotic ribosome has small (40S) subunit and large (60S) subunit. Each subunit exists separately in the cytoplasm and they join together on the mRNA during translation. Ribosome subunits consist of proteins and ribosomal RNA (type of non-coding RNA) In translation, the codons of mRNA are read from 5' to 3' direction by molecules called transfer RNAs. One end of tRNA has set of 3 nucleotides (anti-codons) that matches with the mRNA codon through base pairing. The other end of tRNA carries a specific amino acid. In prokaryotes, there is an area called Shine-Dalgarno box (ribosome binding site) that interacts with the ribosomal RNA molecules. This interaction anchors the small ribosomal subunit to the correct mRNA location. The initiator tRNA carrying the methionine binds to start codon (AUG) of mRNA at the ribosome's P-site. Then large ribosomal subunit joins the complex. In eukaryotes, instead of bindin to the Shine-Dalgarno sequence, the eukaryotic initiation complex (tRNA and 40S ribosomal subunit) recognises 7-methylguanosine cap at the 5' end of mRNA. Ribosome moves to the 5' Cap with the help of Cap-binding protein and initiation factors. Once the ribosome at the 5' cap, it scans the mRNA in the 5' to 3' direction, searching for the AUG start codon. According to Kozak's rule, the nucleotides around the start codon indicates whether it is the correct start codon for initiating translation. The basics of elongation are the same in both prokaryotes and eukaryotes. Translation termination occurs when a stop codon (UAG, UAA, UGA) is encountered on the mRNA. Release factors, that resemble tRNA, recognises these stop codons and instruct peptidyl transferase enzyme to add a water molecule to the carboxyl end of P-site amino acid This reaction forces the P-site amino acid to be released from its tRNA and newly formed polypeptide chain is also released from the complex.
